EXPERIMENTS WITH ENZYMES - TRYPSIN


Reagents

  • Casein or fibrin threads
  • Trypsin solution
  • Sodium carbonate, 1% solution (1.0g sodium carbonate in 100ml of water)
  • Ninhydrin solution (1%)

Procedure

  • Mix the fibrin threads with 2 ml trypsin solution and 2ml of 1% sodium carbonate solution.
  • Adjust the pH between 7.0 and 9.0 with sodium carbonate or NaOH solution.
  • Incubate the mixture at 37°C for 30 minutes. Observe the dissolution of fibrin threads.
  • The end product, peptides can be tested by the development of pink colour by the addition of 1% Ninhydrin solution.
