BACTERIAL COUNT - PROCEDURE

  • Prepare Strained Rumen Liquor (SRL) by filtering the rumen liquor through four layers of muslin cloth.
  • Draw this SRL up to mark 0.5 in the R.B.C pipette.
  • Carefully draw crystal violet stain solution up to 101 mark without any air bubbles.
  • Mix the SRL sample and stain solution by rotating the RBC pipette horizontally between the two palms.
  • Discard 3 or 4 drops of the unmixed stain solution from the stem of the pipette.
  • Focus Neubaur counting chamber covered with cover slip under high power.
  • Charge the diluted SRL between the counting chamber and the cover slip.
  • Keep it undisturbed for few minutes. Count the number of bacteria in five medium squares like R.B.C. counting.
