 i. Deproteinization of serum

  • Pipette into a clean dry test tube labeled (T) as follows

  • Mix well and let stand for 10 minutes at room temperature. Centrifuge at 3000 rpm for 5 minutes.

ii. Colour development

  • Pipette into clean, dry test tubes labelled blank (B), standard(S) and Test(T) as follows

  • Mix well and let stand in the dark at room temperature for 15 minutes. then measure the absorbance of blank(B), standard(S) and test(T) against the distilled water at 710 nm or with a red filter in a photoelectric colorimeter.
