SIMPLE STAINING

Loeffler's alkaline methylene blue

  • This stain is particularly useful to show McFadyean's reaction with Bacillus anthracis. This is also used to demonstrate metachromatic granules in case of Corynebacterium and as a counter stain with Ziehl- Neelsen's stain for acid-fast organisms.

Procedure

  • Prepare a smear and fix it on the flame.
  • Pour few drops of simple stain over the smear and allow it 1-2 minutes.
  • Wash the smear in tap water and air dry it.
  • Examine under oil immersion of microscope.
