The queen is inseminated vaginally with a minimum of 5×106 spermatozoa.

The insemination pipette is passed through the vagina to the area of the cervix, and the insemination dose is expelled.

The time of breeding coincides with the maximum hypertrophy of the vaginal epithelial cells.

Consequently, a vaginal wash is performed to check that the queen is in estrus prior to insemination.
After insemination, the queen must be given LH or human chorionic gonadotrophin to induce ovulation.
