This is a very old method of semen preservation.

In areas where refrigeration facility is poor, semen may be stored at ambient temperature.

The keeping quality of semen is poor at room temperature due to the faster utilization of nutrients by the spermatozoa and also due to bacterial multiplication.

So obviously it is mandate to add sufficient antibiotic to control the multiplication of bacteria.

Various workers have evolved different extenders for the preservation of semen at room temperature (18 – 25oC). Some of them are as follows.
