Procedure

  • Place a small drop of nigrosin close to one end of a clean slide
  • Using a sterile technique, place a loopful of culture inoculum (from the culture medium) in the drop of nigrosin and emulsify (mix) well
  • With the edge of a second slide held at 30º angle and placed on the bacterial – dye suspension on the first slide, push the suspension over the slide with a rapid and smooth motion to form a thin smear.
  • Air dry; do not heat fix the smear on the slide
  • Examine the slide under oil immersion objective
  • Observe the morphological characteristics of bacteria and record the results
