Method

Fill culture tubes with glucose broth or TSB. Prepare five sets of five tubes and label them with the name of the organism and the lethal temperature. Inoculate the tubes with respective organism and heat the tubes in water bath at 60º, 70º, 80º, 90ºC and 100ºC for 10min each. Remove from water bath and cool by immersing in cold water and immediately incubate at 37ºC. |
