Method
Add 0.1g of soil/sediment sample 0.1ml of pond water to nitrite formation medium (adjust pH to neutral of slightly alkaline) and incubate at 25ºC for seven days. Test the culture for presence of nitrites by adding three drops of Trommsdorf solution with one drop of sulphuric acid to a drop of culture. Positive test is indicated by the appearance of intense blue-black colour. Test for ammonia with Nessler’s reagent. Negative result for ammonia indicates complete oxidation of ammonia to nitrites. Isolate and develop pure cultures. Make a gram-stain preparation to observe morphology. |
