PROCEDURE

  • Keep the slide over a staining rack and flood the smear with crystal violet and allow it to react for 1-2 minutes.
  • Wash the smear with running tap water.
  • Flood the smear with Gram’s iodine and allow to react for one minute.
  • Wash the smear with running tap water.
  • Decolorise the smear using 95% ethanol for 10-20 seconds.
  • Wash the smear with running tap water.
  • Counter stain with safranin for 1-2 minutes.
  • Wash the smear with running tap water.
  • Blot dry and focus the smear under low power, high dry and oil immersion.
