PRINCIPLE

  • In this test the antigen and antibodies are incubated in the presence of guinea pig serum which acts as a source of complement. Then the indicator system consisting of antibody coated sheep RBC. When antigen and antibody are present in the test system they fix the complement and not available to lyse the added  sheep RBC. When antigen or antibody is lacking in the test system, the complement is available to lyse the sensitized SRBC as the complement is not fixed.
  • The test can be performed in small test tubes (Wasserman’s tubes), haemagglutination plates or micro titration plates.
