Reagents

  1. Lipase substrate: Olive oil, 50% (v/v) and sodium azide, 0.1% added as preservative. Shake vigorously for a minimum of 20 sec. before using. Store at room temperature (18- 26ºC). Do not freeze.
  2. Trizma buffer (Tris (hydroxymethyl) amino methane) 0.2 mol. pH 8.0 at 25ºC: Add 0.1% sodium azide as preservative. Store at room temperature (18-26ºC). Discard if turbidity develops.
  3. Thymolphthalein indicator: Thymolphthalein 0.9% (W/V) in ethanol. Store tightly capped at room temperature (18-26ºC).
  4. Ethanol : 95%
  5. Sodium hydroxide 0.05 N: Prepare 0.1NaOH solution and standardize by titration against a standard 0.1 acid. Using the formula V1 N1 =V2 N2 find out the normality.  
Then prepare exactly 0.05 N NaOH solution. Store tightly capped in pyrex or polyethylene bottle.
