Analysis for markers

  • Resuspend the pellets individually in an appropriate volume of homogenisation buffer
  • Analyse and detect the presence of DNA, succinate dehydrogenase, acid phosphatase, catalase, glucose -6-phosphatase, RNA, and glucose 6 phosphate dehydrogenase.
  • These substances serve as the markers to help detect the presence of nucleus, mitochondria, lysosomes, peroxysomes, endoplasmic reticulum, rough ER with ribosomes) and cytosol
  • Analyse redissolved pellets to detect, the presence of these subcellular components. Positive results when obtain indicate the presence of specific organelles in the pellet formed at each step of differential centrifugation.

Note:

1. Maintain a temperature of 0-4ÂșC at all stages of the experiment.

2. Follow the protocols given in experiments to assay the marker substances/enzymes mentioned in this experiment
