5.3.2.5. Detection
After the unbound probes are washed away, the blot is ready for detection of the probes that are labeled and bound to the protein of interest. There are several detection methods and the most common detection method is enzyme linked secondary antibody. When the enzyme converts the substrate into an insoluble colored product, it gets precipitated onto the nitrocellulose membrane. The presence of colored band indicates the position of the protein of interest. The protein can be identified by comparison of the blot with the stained gel of the same sample. |
