TOTAL PROTOZOAL COUNT - PROCEDURE

• Prepare Strained Rumen Liquor (SRL) by filtering the rumen liquor through four layers of muslin cloth.
• Dilute 5 ml of SRL with 25 ml of 10% Formal saline.
• Take 5 ml of this diluted SRL and add 5 drops of 2% Eosin solution.
• Focus the chamber under high power and charge the diluted and strained SRL into the Neubaur counting chamber without air bubble.
• Keep it undisturbed for few minutes. Count the number of protozoa in four W.B.C. squares.
• As the protozoal populations tend to vary based on the geographical location which the animal is reared, type of diet being fed, age and other factors, the pictures provided in this lesson facilitate the learners for gross identification of protozoa.
