PRINCIPLE 

  • Passive haemagglutination involves the sensitization of certain mammalian RBCs with soluble antigens of microorganisms and subsequent agglutination of these sensitized cells by specific antisera directed against antigens of the microorganisms.
  • The erythrocytes merely serve as indicator of antigen and antibody reactions. The antigen is called erythrocyte sensitizing substance.Polysaccaride antigens get absorbed on sheep erythrocytes very easily. But effective coating of protein antigens, the cells require to be penetrated with certain reagents.
  • The commonly used reagents are tannic acid, glutaraldehyde, periodite, pepsin, trypsin, Bis-diazotozed benzidine, chromic chloride.
