PREPARATION OF E.COLI COMPETENT CELLS

• The E.coli cells picked from a single colony is gorwn freshly until it reaches an OD of 1.6 -1.8 (less than 10⁸ cells / ml).
• The E.coli cells then made competent by CaCl2 treatment
• The prepared competent cells are aliquoted and stored at -80⁰ C until use
