Biochemical Techniques and Instrumentation

Reagents

1. Denaturation Solution : 1.5 M NaCl and.0.5 M NaOH. Store at room temperature (RT)
2. Renaturation Solution : 1.0 M Tris.Cl (pH 8.0) and 1.5 M NaCl. Store R.T.
3. 20X SSC (Saline sodium citrate) buffer : 175.3 g NaCl, 88.2 g Sodium Citrate (trisodium salt), pH to 7, Distilled water to 1 L
4. Ethidium Bromide : 10 mg/ml in water. Stir 6-12 h. Store 4°C in brown bottle.
5. 50X TAE (Tris acetate EDTA) buffer: Trisma base. 242 g/L, Glacial Acetic Acid: 57.1 ml/L, O.5 M EDTA (pH 8.0): Prepare 100 ml and store R.T.
6. 10X Loading Buffer : 0.42 % Bromophenal blue, 0.42 % xylene cyanol, 50% glycerol in water. Store at 4°C
7. Size Markers : 50 m g/ml HindIII to cut mlDNA in T.E. Store -20°C
   50 ug/ml HaeIII to cut rX174 DNA in T.E.
8. 1/10 volume 10X loading buffer
