Principle

In the agar gel immunodiffusion test antigen and homologous antibody placed in wells cut in inert agar diffuse towards each other and precipitate to form an opaque line in the region where they meet in optimal proportions. In the optimal antigen antibody proportion, a chain (lattice) of Ag and Ab thus forms and becomes visible. The agar gel immunodiffusion test is influenced by electrolyte pH, temperature and time.
