PRINCIPLE

  • Glucose is oxidized by the enzyme glucose oxidase (GOD) to give D-gluconic acid and hydrogen Peroxide (H2O2).
  • Hydrogen peroxide in presence of enzyme peroxidase (POD) oxidizes phenol, which combines with 4-aminoantipyrine to produce a red colored quinoneimine dye.
    The intensity of the colour developed is proportional to the glucose concentration in the sample.


    D-Glucose + H2O → D-Gluconic acid + H2O2
    H2O2+ 4-amino + Phenolinoantipyrine + phenol → Red quinoneimine dye + H2O
