Principles of Biochemistry (2+1)

Procedure

• Weigh accurately 10g of the minced sample.
• Homogenise with a mixture of 20ml of chloroform and 10ml of methanol.
• Add 10ml of chloroform to the above mixture and blend for 30 seconds.
• Add 10ml of distilled water and filter the homogenate through Whatman No.1 filter paper.
• Transfer the filtrate into a separating funnel and allow to separate
• Separate the chloroform layer into a previously weighed beaker.
• Evaporate over a water bath to remove chloroform from the beaker.
• Dry the beaker in a hot air oven at 100^ºC for 30 minutes and weigh.
