Fixation

  • It is the first step in processing of tissue for permanent microscopic preparation.
  • The fixation process should preserve the cells in fresh condition as possible.
  • During fixation the gross physical structure and cellular details of the material are pressurised.
  • Large volume of fixative should be used.
  • Tissues should be isolated and kept in physiological saline before fixing it.
  • Selected tissue should be kept immersed in the fixative such as Bouins fluid for 3-12 hours or even 24 hours depending on the nature of the sample tissue for effective fixation. After wanely, it should be transformed into 70% above alcohal.
