VMC 211: General Veterinary Microbiology (1+1)

PROCEDURE

• Place a coverslip over a flat surface and put small amount of petroleum jelly on the four corners of the coverslip.
• Place a small drop of liquid medium containing bacteria at the centre of the coverslip. The drop should be very small otherwise it will get crushed when the cavity slide os placed.
• Place a cavity slide over the coverslip in such a way that the concave surface is over the drop.
• Gently reverse the slide. While reversing care should be taken not to disturb the hanging drop.
• First focus the slide under low power and observe the edge of the drop.
• Change the power to high dry and observe the bacterial motility at the edge of the drop.
