Procedure

Cut a well pattern as given in the template. The distance between the wells should be 4mm. Remove agarose from the well using the needle and place them on the electrophoresis tank.

Fill the buffer tanks with barbitone or Manthoney's buffer.

Link each end of the slide to the electric circuit by filter paper strips pre soaked in barbitone or Manthoney's buffer.

Wells towards cathode are filled with antigens (known +ve, known –ve and test antigen) and towards anode are filled with antiserum.

Electrophoresis is carried out using a current of 8 milliamperes / slide for 45-60 minutes.

Switch off and examine for white lines of precipitation between the pairs of wells. Readings are facilitated by oblique light.

Fix the protein by immersing the slide in 10% glacial acetic acid.

Stain the slide and observe for the presence of precipitation line.
