PRINCIPLE

Principle of estimation  of ALT (SGPT) (Reitman and Frankel’s method)

• Serum Glutamate Pyruvic Transaminase (SGPT) or Alanine amino transferase (ALT) catalyses the following reaction:      

µ- Ketoglutarate + L-Alanine → L-Glutamate + Pyruvate

• Pyruvate so formed is coupled with 2, 4 – dinitrophenyl hydrazine (2,4 – DNPH) to give the corresponding hydrazone, which gives brown color in alkaline medium and this, can be measured colorimetrically.
