STAINING OF PRECIPITATION BANDS

Staining solution

• Coomassie brilliant blue (R 250) - 0.5 g
• Ethanol (96%) - 45.0 ml
• Glacial acetic acid - 10.0 ml
• Distilled water - 45.0 ml

Destaining solution

• Staining solution without Coomassie brilliant blue.

Washing and drying

• The plates are submerged in normal saline with several hourly changes for 24h in order to elute all non precipitated (free) proteins.
• Keep the slides in distilled/tap water for one hour for removing salt.
• Finally dry the gels overnight at 37⁰C by covering the slides with a wet filter paper.
• After drying remove the filter paper.

Staining

• Stain the slides with staining solution for 15 –30mts.
• Destain the slide with destaining solution for 5 –10min.
