Spot Test

  • This test is used for rapid detection of the presence of a haemaggglutinating virus. Mixing two or three drops of suspected sample with equal quantity of 5% chicken red blood cells in normal saline. The appearance of the clumps of agglutinated red cells, indicate the presence of a haemagglutinating virus in the sample.

Micro Haemagglutination Test

  • Dispense 25 µl of Phosphate buffer saline / normal saline into (one row for each sample) twelve wells of the plate.
  • Add 25 µl of sample into the first well.
  • Use a multichannel pipette to make two-fold serial dilutions along the row until the second last well from the end.
  • The last well is the RBC control. Do not dilute this well.
  • Gently tap the sides of the microwell plates to mix the reagents.
  • Add 25 µl of 1 percent washed red blood cells to each well including the control wells in the last column.
  • Gently tap the sides of the microwell plates to mix the reagents. Cover the plates with a lid. Allow to stand at room temperature for 30 -40 minutes.
  • Observe and record the results.
