Principle

The estimation involves extraction of histamine by perchloric acid, followed by n-butanol under alkaline condition and finally collecting it in acid- aqueous phase. The acid-aqueous phase is then condensed with o-opthalaldehyde to make them fluorescence. The fluorescence is is read at an excitation wavelength of 340 nm and emission wavelength of 425 nm in a spectrofluorometer.
