Estimation of protein by Lowry's Method

Principle

  • Protein reacts with Folin-Ciocalteau reagent to give a coloured complex.
  • The colour so formed is due to the reaction of the alkaline copper with protein and the reduction of phosphomolybdate by, tyrosine and tryptophan, the aromatic amino acids, present in the protein.
  • The intensity of the colour formed is directly proportional to the amount of these compounds present in the protein.
  • The colour developed is measured at 660nm in a spectrophotometer.
