Physiology of Finfish and Shellfish (2+1)

Procedure

Take liver or muscle or any other tissue (if fresh not more than 20-30 mg) from the animal provided and grind well with mortar and pestle with 1 ml of distilled water. To this solution add 2 ml of biuret reagent and transfer into a test tube. Wash the mortar with 2 more ml of the biurette reagent and add it to the original mixture. Wait for some time (30-60 minutes) and filter the solution with the help of a filter pump using a glass filter paper.

Prepare a blank solution taking 1 ml of fistilled water and 4 ml of biurette reagent. Read the intensity of the colour in a Spectrophotometer at 540 m. Determine the optical density and read the quantity of protein from the standard graph.
