HAEMATOCRIT METHOD

Procedure

  • Fill a heparinized microhaematocrit tube with blood and centrifuge for 1 minute at 1500 rpm.
  • Place the tube under a dissecting microscope and examine the interface of the plasma and buffy coat for motility. Microfilariae (and haemoflagellates) will be observed actively swimming in the plasma.
  • Break the tube at the buffy coat interface and dab the buffy coat and a small amount of plasma on a slide; add a coverslip and examine for live microfilariae (at 10X) or allow it to air dry, fix for 1 min with methyl alcohol and stain with Giemsa for 30 min for identification (1.5 ml Giemsa’s stain in 15 ml distilled water buffered to pH 7.2).

*** Note that heparinized and non-fixed (non-formalin treated) blood specimens should be examined on the day of collection.
