'O' antigen

‘O’ ANTIGEN

  • A smooth colony of a Salmonella culture is selected and inoculated into 10ml broth and incubated at 37 0 C for 6 hours.
  • Three or four agar plates were inoculated with the 6 hour old culture and incubated at 37 0 C for 18 hours.
  • The bacterial growth was harvested with saline and mixed thoroughly.
  • The bacterial suspension was heated for 30 min in flowing steam (1000C) .
  • Formalin 0.5% is added to the antigens as preservative.
  • The antigen may be stored in the refrigerator.
