VMC 221: Veterinary Immunology and Serology (1+1)

H - ANTIGEN

• Passage Salmonella culture (flagellated) into Cagie’s tube 2 or 3 times for enhancing its motility.
• Inoculate the culture into broth (equal mixture of trypticase soy and tryptose broth contained in 10 ml amount in large tubes (18 x 140 to 150mm) and incubate for 18 hours at 37⁰C.
• Add equal volume of formalinized saline solution ( 0.85% sodium chloride + 0.6% formalin into equal volume of the culture and incubate at room temperature for at least 1 hour.)
• Store in the refrigerator overnight before use.
