Procedure for preparation of O antigen from bacterial suspensions treated with alcohol

PROCEDURE FOR PREPARATION OF "O" ANTIGEN FORM BACTERIAL SUSPENSIONS TREATED WITH ALCOHOL

  • Bacterial suspensions treated with alcohol give excellent results and the procedure of their preparation is as follows
    • The bacterial culture grown on agar plate for 20 hours at 370C.
    • The bacterial growth is harvested with saline and a desire saline suspension is made.
    • One volume of 96% alcohol equal to that of the saline used is added and mixture incubated 370C for 20 hours.
    • The dense suspension (stock alcohol- antigen) is stored in refrigerator where it will retain its properties for many years.
