MECHANISM OF TOXICITY

  • These anticoagulants have basic coumarin or indanidone nucleus.
  • They antagonize vitamin K, which in turn interferes with the normal synthesis of anticoagulant proteins (Factors II, VII, IX and X) in the liver. So prothrombin conversion to thrombin is affected.
  • A latent period which depends on species, dose and activity is required for using up the already present factors.
