Procedure

The test should be carried out between 200C and 250C. (At higher temperature false positive reactions caused by Auto - Agglutination may be observed).

Place a drop of suspected serum on a glass slide/plate.

Place a drop of colored antigen next to the serum sample.

The drops are then mixed gently with stirring-rod and spread to a circular area of approximately 1.5cm diameter.

After a few minutes, in case of positive reaction ,clumping takes place (Agglutination).

Confirm the reaction with suitable positive and negative controls.

Appearance of medium sized particles within 2-3 minutes

Appearance of very tiny powdery paticles developing in 4- 5 minutes

Negative
|
Absence of agglutination.
