PROCEDURE

  • The test should be carried out between 200C and 250C. (At higher temperature false positive reactions caused by Auto - Agglutination may be observed).
  • Place a drop of suspected serum on a glass slide/plate.
  • Place a drop of colored antigen next to the serum sample.
  • The drops are then mixed gently with stirring-rod and spread to a circular area of approximately 1.5cm diameter.
  • After a few minutes, in case of positive reaction ,clumping takes place (Agglutination).
  • Confirm the reaction with suitable positive and negative controls.

Analysis

+++   A strong positive reaction visible within 3 seconds
++   Appearance of medium sized particles within 2-3 
  minutes
+   Appearance of very tiny powdery paticles 
  developing in 4- 5 minutes
Negative   Absence of agglutination.
