Principle

  • SDS is an anionic detergent, which binds strongly to and denatures protein.
  • The number of SDS molecules bound to a polypeptide chain is approximately half the number of amino acid residues in that chain.
  • The protein–SDS complex carries net negative charges, hence move towards the anode when electricity is applied and the separation is based on the size of the protein.
