Precautions in the preparation of semen diluents

Only double glass distilled water, fresh eggs and pure chemicals are used for preparing diluents.

Care is taken not to rupture the yolk membrane before it becomes free of white.

The yolk membrane should be retained with filter paper and discarded.

Enough buffer solution is prepared in bulk, sterilised and kept in refrigerator.

Diluents are prepared on the previous day itself of collection and kept in a refrigerator until used before dilution, the temperature of the diluent should be 30 ° C and this is achieved by keeping it in a water bath for 15-20 minutes before dilution.

The semen is always kept at 30 ° C during evaluation.

Dilution should be done within 10 minutes after collection.

Glycerol when added should be warmed to 40 ° C. After addition, the glycerol should be mixed well with the diluents by stirring gently with a sterile glass rod for 20 minutes.

Always use sterilised glassware during the entire operation.
